Illustrated here is the ZnT8 protein, a novel target of T1D autoimmunity. It is a beta cell specific zinc transporter localized on the membrane of insulin secretory granules. ZnT8 contains six transmembrane domains and a histidine-rich loop between transmembrane domains 4 and 5, which is the putative zinc binding domain. The divalent zinc cation would be transported into the lumen of the insulin secretory vesicle where it would bind to insulin and create the zinc-dependent hexameric complex.
